Fully Automated, One Hour Sample Preparation for Ultrafast N-Glycosylation Analysis of Antibody Therapeutics

Abstract

SCIEX has recently introduced an easy-to-use magnetic bead based sample preparation workflow, enabling the preparation of up to 96 samples in one hour. This novel protocol aims to alleviate the hard-to-automate centrifugation and vacuum-centrifugation steps used in other N-glycosylation analysis methods. In this presentation, we demonstrate the next level of this work by fully automating all steps of the optimized magnetic bead based method to eliminate manual sample handling. The one-hour sample preparation protocol for CE-LIF based N-glycosylation analysis of antibody therapeutics includes endoglycosidase digestion, fluorophore labeling and sample clean-up. Automated liquid handling offers fast and precise sample preparation with minimized contamination risks and increased reproducibility to streamline laboratory operations. This rapid sample prep allows you to scale up from single vial analyses to 96 well plates for high throughput screening by using a Laboratory Automation Workstation (Beckman Coulter Biomek FXP®) to accomplish complete sample processing in 60 minutes.
