Abstract
High pressure freezing offers versatile and deep capability in the study of intricate changes in fine structure or cellular dynamics. From the internals of a single cell to complex layers within small whole organisms, the EM ICE occupies a key point in many electron microscopy (EM) workflows. These workflows extend from correlative light and electron microscopy (CLEM) methods to straightforward freeze substitution by resin embedding for ultramicrotomy.

This webinar is an overview of the various workflows enabled by high pressure freezing on the EM ICE.
- Live cell CLEM for stabilizing transient and rare biological processes
- In situ light and electrical stimulation of live cells, with millisecond timing precision relative to cryofixation
- Freeze substitution for room temperature EM
- Freeze fracture to expose the internal structure of cells
- Lamella milling by focused ion beam
- Cryo ultramicrotomy, for cryo transmission EM of thin sections

The EM ICE, combining superior high pressure freezing with support for a wide variety of workflows, is an excellent platform for more than just single project.
