Abstract

Cell counting is a fundamental step in many workflows, including cell culture, cell analysis, transfection, single cell sequencing, and cell therapy. Automated cell counters have improved the speed, accuracy, traceability, and complexity of the data acquired. Accurate cell counts depend on sample preparation, reagent selection, and counting method.  This presentation will cover cell counting techniques using the Invitrogen™ Countess™ 3 and Countess™ 3 FL automated cell counters, offering tips for increased accuracy and details on nuclei counting for single-cell analysis workflows.

Countess 3™ automated cell counters utilize artificial intelligence (AI), enabling users to quickly and easily obtain accurate and precise counts, concentrations, viability measurements and/or fluorescent expression of their cells for a variety of downstream applications. The Countess™ 3 FL provides two count methods (brightfield-based or fluorescent-based) allowing users to obtain counts specifically for their cells of interest. The fluorescent-based counting algorithm was specifically developed for single-cell analysis workflows and enable researchers to confidently count cells or isolated nuclei for downstream use in single-cell RNA sequencing (scRNA-Seq) and single-cell sequencing assays for transposase-accessible chromatin (scATAC-Seq).
