Abstract
AAVs are attractive viral vectors for gene therapy due to their lack of pathogenicity, low immunogenicity, broad tropism and persistent transgene expression in both proliferating and quiescent cells. Both the purity of AAV capsid proteins and AAV genome integrity analysis are of significant importance. The purity of AAV capsid proteins can affect vector efficacy, while the analysis of AAV genome integrity provides insight into transgene integrity and ensures product safety and potency.
 
In this webinar, Tingting Li will illustrate how multi-capillary electrophoresis can be used to accelerate AAV protein purity method optimization analysis up to 12x faster than single-capillary methods, while maintaining consistent, highly sensitive results. She will also present direct comparison of genome integrity analysis on a single capillary and multi-capillary CE-LIF system. This analysis was performed, showing similar data quality and consistent corrected peak area percentage of the genome components between the two systems. With the multiplexing capability, quick assessment of the genome integrity of AAVs from different serotypes or different genome payloads can be achieved for up to 8 samples in a single run.
