Abstract

Genotyping assays are essential in a variety of plant and animal applications including Marker Assisted Selection (MAS) and Quantitative Trait Locus (QTL) Mapping. While microarrays and endpoint PCR assays have been traditionally employed for these purposes, these methods lack contextual data of surrounding genomic regions, are limited to single nucleotide variants, and produce data with high false-positive calls. Target enrichment combined with next-generation sequencing is a compelling alternative that provides more complete genomic information of subsets of marker loci along with the ability to discover newly introduced variants. However, the high-costs and limited sample throughput of existing target enrichment strategies have rendered next-generation sequencing approaches largely impractical for genotyping workflows.

 

During this webinar, we will discuss the development and verification of the automated NEBNext DirectÒ Genotyping Solution on the Beckman Coulter i7 workstation. This approach combines innovative molecular biology with user-friendly, liquid handling automation to achieve highly-multiplexed sample processing. The result is a robust and cost-effective targeted NGS method capable of meeting the growing demands of plant and animal genotyping applications.

  • Genotyping data demonstrates robust results across 96 samples, with improvements in several key metrics over manual processing.
