Abstract
The ability to synthesize and sequence vast numbers of DNA constructs is a cornerstone of modern high-throughput biological screening and discovery. As a result, the need for faster, easier, and more efficient sample and library prep methods has become increasingly important. This webinar will explore new methods and technologies to accelerate the sequence-based screening of plasmids for a wide range of needs in discovery, research, and production-scale biotechnology. The presentation will highlight the development of seqWell’s ExpressPlex technology and explore how this workflow has been combined and optimized with rolling-circle amplification (RCA) at Octant toward the development of their upcoming high-throughput plasmid sequencing pipeline update, OCTOPUS v3.0–a tool which has been instrumental in facilitating Octant’s internal synthetic biology and drug discovery processes. The products and processes outlined in this discussion can be easily implemented by interested users to significantly improve the pace and scale of synthetic construct and plasmid screening and sequencing.

Learning Objectives
  • Review how rapid NGS of plasmid and amplicon libraries can drive efficiency gains in synthetic biology R&D and the therapeutic discovery process
  • Recognize new methods of sample and library prep that will reduce labor, time, and consumables
  • Enable additional strategies for creating flexible, high-throughput workflows
