A physiologically-relevant 3D ECM for in vitro oncology research and intelligent high-content imaging of 3D models

Trends continue towards the development of more sophisticated and physiologically relevant 3D in vitro models, such as stem cell-derived models, organoids, organ-on-a-chip, and cells in extracellular matrices. In parallel, fully automated imaging solutions now enable an ever-deeper insight into cell compartments while intelligent software tools are turning the high information content of microscopy images into meaningful results.

In this webinar, HCS Pharma scientists will present a patented 3D cell culture system, BIOMIMESYS®, which mimics the key features of the cellular microenvironment, either physiological or pathological, in a ready-to-use format. This unique 3D extracellular matrix (ECM) is different from standard hydrogels used for 3D cell culture as it is made of native ECM components: Hyaluronic Acid (HA) combined with collagens, adhesion peptides, in the form of a unique hydroscaffold which exhibits dual behavior between hydrogels and solid scaffolds. Solid tumors are often associated with a modified microenvironment, with an enhanced collagen content and increased stiffness. These ECM changes are now recognized as important drivers of cancer initiation and progression, and also impact the treatment of cancer lesions. The BIOMIMESYS® Oncology matrix allows cancer cells to be cultured in a tumor-like environment, with the possibility to tune the composition and stiffness of the matrix. We will show how this 3D model can be used in cancer research through results obtained by confocal imaging and subsequent image analysis.

We will also present our next generation of automated imaging system, the ImageXpress Confocal HT.ai, which brings best in class hardware flexibility and throughput as well as machine learning capabilities to translate complex 2D or 3D images into scientific discovery.
