Although the classical second-generation CAR design has been shown to be very effective when used to treat hematological malignancies, this traditional CAR design has failed to promote equally significant antitumor activity in non-hematological cancers. Killer immunoglobulin-like receptors (KIRs) are an important family of immune receptors used by T cells and NK cells. Considering that their multi-chain design may have the advantage of evolutionary selection, Creative Biolabs designed a more realistic CAR design based on unique KIRs found in humans and non-human primates. This KIR-based CAR (KIR-CAR) naturally interacts with the DAP12 protein, delivering important signals to T cells that promote T cell proliferation, cytokine secretion, and cytotoxicity.
