    Biography
      Wieland Keilholz received a Ph.D. in Chemistry from the University of Heidelberg in collaboration with the German Cancer Research Center and the University of Tübingen. As a post doc, he implemented and ran a mass spectrometry service at the University of Tübingen. He then transferred from research to customer facing roles, holding positions with Affymetrix and NuGEN Technologies before joining BD Biosciences in 2016 where he was heading the European Field Applications Support team for the BD Rhapsody product portfolio. His current role as single cell Solutions Marketing Development Manager is focusing on running collaborations to further develop the BD ONE solution approach.

      Biography

        Pawel Zajac is the senior cell and molecular biology marketing manager in Europe, Middle East and Africa at Illumina. In this role, he is responsible for developing and executing the marketing plan for this segment. His prior roles at Illumina span several functions in sales and marketing. Pawel has a PhD from the Royal Institute of Technology (KTH) in Stockholm and worked as a PostDoc at the Karolinska Institute in Stockholm. His research focused on method development in single-cell RNA sequencing and multiplex amplification. Pawel is the author of >10 peer-reviewed articles in journals including Nature Methods and Genome Biology.

        Biography
          Tilmann is a biochemist by training. After a PhD at the Robert-Koch-Institute in Berlin and a postdoctoral stay with Giulio Superti-Furga, he joined Haplogen as Principal Scientist and, later, Head of Research. With the advent of CRISPR, Tilmann focused his team on CRISPR gene editing and quickly produced the largest collection of isogenic human knockout cell lines available to date. This became the basis for the commercial success of Haplogen Genomics and its acquisition by Horizon Discovery. At Horizon, Tilmann served as R&D Director and Head of Innovation with global oversight of all R&D projects. In early 2018, Tilmann founded Aelian Biotechnology, where he currently serves as CSO. He is passionate about science and enjoys working with multi-disciplinary and multi-national teams.

        CRISPR screening has become the prime discovery tool in modern biomedical research and drug discovery. At the same time, most screens conducted today rely on rather simplistic read-outs.
         
        At Aelian, we use the CROP-seq (CRISPR droplet sequencing) workflow developed by Aelian co-founder Christoph Bock to combine CRISPR screening with single-cell transcriptomics, leveraging two transformative technologies to enable genetic screening for complex phenotypes. Our powerful approach has broad applications in identifying novel drug targets or elucidating unknown mechanisms of actions of drugs.
         
        Currently, most single-cell RNA sequencing platforms utilize an unbiased sequencing approach. While powerful, it typically delivers shallow sequencing data for each single cell. At Aelian, we combine CRISPR perturbation with targeted single-cell RNA sequencing using the BD Rhapsody workflow, coupled with Illumina sequencing readout, to focus sequencing resources on the most informative transcripts. The data we gathered suggest that targeted sequencing leads to a reduction in cost as cells can be sequenced at 10x reduced depth. At the same time, the signal-to-noise ratio improves significantly, thus enhancing the dynamic range of CROP-seq screens.
         
        Overall, advances presented here will pave the way towards genome-scale single-cell CRISPR screens.
