Abstract

High-parameter (greater than 20-color) flow cytometry is increasingly adopted in clinical settings. It provides unparalleled capabilities for immunophenotypic characterization in the diagnosis of leukemia and lymphoma. However, it also significantly increases the complexity of the bench workflow, data analysis, and diagnostic report. This presentation details the development and implementation of a 30-color flow cytometry panel, sharing critical practical insights gained from our institutional experience, with particular emphasis on three critical areas: method optimization, data preprocessing, and automated data analysis.

Learning Objectives:

1. Describe the advantages and challenges of implementing high-parameter (≥20-color) flow cytometry in clinical diagnostics.

2. Explain key strategies for method optimization, data preprocessing, and automated data analysis in high-parameter assays.

3. Discuss practical considerations for integrating high-parameter flow cytometry into clinical workflows to improve diagnostic accuracy and efficiency.
